Effect of collection time on pollen viability and meiotic behavior of corn (Zea mays L.)

Pollen viability studies are determining factor for the success of breeding and genetic resources conservation programs of a species, since it is essential that genotypes have a high percentage of viable gametes for the fertilization process to occur efficiently. In this sense, the aim of this work was to evaluate the reproductive aspects of the simple hybrid 2B810 PW (Zea mays L.), through meiotic behavior and pollen viability at nine different collection times. The botanical material was grown in a sprinkler irrigated outdoor system, between the months of July and October 2018. Male flower buds were collected in different sizes and stages of development, from plants randomly selected in the field, starting at 06: 00hs and finish at 14: 00hs. For the study of pollen viability, the following dyes were used: Alexander Triple Solution (STA) and Acetic Carmine (CA). Through statistical analysis it was possible to verify that, from 7 am to 1 pm, the viability of the male gametes remained constant, therefore, for the hours from 6 am to 2 pm, it was observed less pollen viability. An average meiotic index (IME) value of 73.7% was obtained and based on colorimetric tests, an average pollen viability of 96.11% was observed. It was possible to conclude that the collection times, evaluated in the present experiment, significantly influenced the pollen viability of the species and that an irregular meiotic index (IME) rate was verified at different times.
